When Running 200 Miles Is Easier on Your Body Than Running 100

After 200 miles and three straight days of running across the Northern Italian Alps, 50-year-old Gregoire Millet wearily walks across the finish line. The brutal 2012 Tor des G?ants is the second ultramarathon he has run, and by pushing his body well beyond what most would consider the brink, he takes the silver medal. He has been racing for nearly 79 hours, much of it in the rain, and has slept for only 6. He has crossed a seemingly endless stretch of rocky crags and endured 15 miles of elevation change. He is exhausted.

Yet, according to new research?which Millet himself helped to author?he'd be more exhausted had he run a race half the distance. "Well, after three days you are not completely fresh, you know" he says, "but it seems the exhaustion is not exponential."

Millet's research team, led by Jonas Saugy, a sports physiologist at the University of Lausanne in Switzerland, has tracked the human body's response to the demands of a 200-mile ultramarathon. In a study published today in the journal PLOS ONE, the scientists report that the Tor des G?ants finishers suffered far less muscle damage and fatigue than those who had run an equally mountainous 103-mile race across the French, Italian, and Swiss Alps. "It was a surprise it was such a big difference," Saugy says.

Saugy and his team took blood samples and performed a 20-minute series of tests on their study participants before, during, and after the race. The researchers measured muscle strain in the knee and foot in a custom-built chair with a testing gauge, and recorded the electrical activity of the muscles with electrical stimulation. Their findings: Tor des G?ants racers finished with roughly 30 percent more strength in their lower leg muscles despite running so much farther.

Saugy argues that the more conservative pace?and the eventual surrender to sleep exhaustion?protects an ultramarathoner's muscles. While racers at the 200-mile Tor des G?ants averaged 3.4 mph over the course (a 17.5-minute-mile pace), those who ran only half the distance in the 103-mile race averaged 4.5 mph (13.4-minute miles). Running at a slower pace means running with a softer gait, and, in fact, many of the ultramarathoners spent much of the second half of the race walking. As for sleep, even the top medalists in the Tor des G?ants yielded to sleep exhaustion and all the racers slept an average of 9 hours, usually in 20- or 30-minute naps.

"A shorter distance means higher intensity," says Martin Hoffman, an exercise physiologist at the University of California, Davis, who was not involved with the study. "Any time you do a longer event your intensity has to go down," says Hoffman. But what's unexpected is just how much less the longer event demands from your legs and feet.

There's plenty more to be learned about how the body responds to such a grueling completion?though even getting this much data was a chore. The Tor des G?ants is so difficult that only two thirds of the runners who enter finish it. To complete this study, Saugy and colleagues had to collect enough participants who'd complete the event and agree to the 20-minute midrace examination (the clock didn't stop while the runners stopped to be tested). While Saugy's team collected 25 subjects, 10 dropped out of the race and an additional six wouldn't stop for the midrace test when the time came.

"And to ask people to do something beyond just running the race is a big deal," Hoffman says, "to get as many as they got is pretty remarkable." Still, he says, the data is robust enough to prove the longer-distance racers had lower levels of muscle damage and fatigue.

Previous studies have shown that sleep deprivation doesn't have a direct effect on the muscle strength of the ultramarathon runners. But Saugy's data shows that even brief sleep breaks were nonetheless an important period of rest for the muscles. "The addition of sleep" once the runners have built up a sleep deficit "was the reason for the muscle conservation," Saugy says.

Ultramarathons, still a growing sport, offer researchers a chance to examine the human body under stresses that would be tremendously difficult?and perhaps even unethical?to produce in a lab. "It's a competition where you can push your body to extreme conditions," he says. "You'll never find conditions like these in other competitions."

Rhapsody debuts app for Windows 8 at Microsoft Build 2013

Today during Microsoft's 2013 edition of Build, Rhapsody announced that it's releasing a version of its music app designed for the Windows 8 operating system. As you might expect, the Win8 variant will bring many of the same features found on its iOS and Android counterparts, including the ability for subscribers to create playlists and stream / download songs from Rhapsody's ample library of tunes. There will be some tidbits tailored specifically for Redmond's OS, however, such as a Snap Mode for simple multitasking and an option that allows tracks to be pinned to the Metro-style home screen. The company told us the application will hit the Windows store shortly, so we'll be sure to update this post as soon as we have a link to the download.

Viewing held for 'Sopranos' star Gandolfini in NJ

PARK RIDGE, N.J. (AP) ? A private viewing for James Gandolfini (gan-dahl-FEE'-nee) has ended in a small New Jersey town near where he grew up.

Mourners at Wednesday's invitation-only wake for family and friends left flowers at the Robert Spearing Funeral Home in Park Ridge.

The 51-year-old star of "The Sopranos" grew up in nearby Westwood. He died last week in Italy.

Nurse Robin Caprio wasn't allowed in to the private viewing and stood outside with a photo of herself and Gandolfini taken when his show filmed a scene in Paterson.

Broadway theaters dimmed their marquee lights in memory of Gandolfini, who earned a Tony Award nomination in 2009 for his role in "God of Carnage."

Gandolfini's funeral will be held Thursday at the Cathedral Church of Saint John the Divine in New York City.

Printable travel game | Simple Mom

A few weeks ago our family hit the road as we made the big move from Alberta, Canada to sunny Arizona. Along with packing, cleaning, and the various tasks that come with a move, I knew I?d also need to prepare for the long drive ahead of us.

With an ample snack assortment, ?a handful of dollar store toys and some favorite movies, I was surprised to find that one of the kids? favorite activities on the trip was a simple ?search and find? game that I?d printed out off the internet.

I?ve expanded on the original printout I used and wanted to share this fun little travel game with you in the hopes that it will come in handy on your next family road trip.

You can download the printable game cards to the travel game here. There are four sheets, 48 game cards in total. I printed mine out at home on white cardstock and cut out the cards using a paper trimmer. I added an extra step and used a punch to round the corners of each card but that?s completely optional.sm_travelgame6

Once you?ve printed and cut out your travel game cards, you could have them laminated to make them more durable. The amount of white paper and ink needed to print these is pretty inexpensive, so leaving them as-is is fine, too ? printing out another page or two if they get worn won?t break the bank.

You can store the cards in a ziplock bag or in one of my favorite storage containers for carrying small items like felts, crayons and card games in the car.

What to do with your travel game cards:

Laser guided codes advance single pixel terahertz imaging

June 25, 2013 ? The universe is awash in terahertz (THz) waves, as harmless as they are abundant. But unlike other regions of the electromagnetic spectrum, THz has proven to be extremely difficult to manipulate in order to capture novel images of objects and materials with which these light waves interact.

Most existing THz imaging devices employ prohibitively expensive technology or require several hours and cumbersome manual controls in order to generate a viable image, according to Boston College Professor of Physics Willie J. Padilla.

Padilla and researchers in his lab recently reported a breakthrough in efforts to create accessible and effective THz imaging. Using both optical and electronic controls, the team developed a single-pixel imaging technique that uses a coded aperture to quickly and efficiently manipulate stubborn THz waves, according to a recent report in the journal Optics Express.

In the so-called terahertz gap, a region of wavelengths that falls between microwave and infrared frequencies, conventional electronic sensors and semiconductor devices are ineffective. Some systems capture only a fraction of a scene and the means to tune these THz waves are inefficient. This has fueled the search for new imaging technologies in order to manipulate THz waves.

Efforts to overcome the challenges of mechanics, cost and image clarity are viewed as a crucial step in efforts to tame the THz gap since imaging and sensing at this frequency holds the potential for advances in areas as divergent as chemical fingerprinting, security imaging of hidden weapons, even real-time skin imaging to promote simple detection of skin cancer.

Central to this challenge is the development of a technology to create efficient masks -- similar to the aperture of a camera -- capable of tuning THz radiation in order to produce clear images in just a few seconds.

Padilla and graduate students David Shrekenhamer and Claire M. Watts report their new single pixel imaging method centers on what they describe as a "coded aperture multiplex technique" where a laser beam and electronic signals are used to send a set of instructions to a semiconductor so it can guide the reproduction of the image of an object after THz waves have passed through it.

A digital micro-mirror device encodes the laser beam with instructions that direct certain segments of the silicon mask to react and allow a selected sample of the THz waves to pass freely through, consistent with the image pattern. The combination of optical instructions and the reaction of the semiconductor create a THz spatial light modulator, the team reports. Functioning like the aperture of a conventional camera, the modulator then guides the digital reconstruction of the entire image based on a broad sampling of THz waves that have passed through the object.

The team's experiments found the method could produce masks of varying resolutions, ranging from 63 to 1023 pixels and acquire images at speeds up to .5 Hz, or about 2 seconds. The early findings "demonstrate the viability of obtaining real-time and high-fidelity THz images using an optically controlled SLM with a single pixel detector," the team concluded.

Padilla said the findings have spurred additional research by his lab into ways to further control THz waves, such as by using the intricate patterns of an engineered metamaterial to further manipulate terahertz waves to create images faster and with increased efficiency.

Pokki Partners With Acer To Offer Zynga Games On PC Desktops ...

Pokki is partnering with Acer and Zynga to offer a new game arcade, featuring the full Zynga.com library of games, on new Windows 8 PCs. The arcade, an extension of the Pokki Game Network, will be preloaded on Acer PCs, along with the popular Pokki Start menu for Windows 8 and hundreds of free apps in the Pokki app store.

Pokki, which is made by SweetLabs, previously won over fans with desktop versions of popular apps like Angry Birds, Pandora and Pinterest, an app recommendation system, and a start menu for Windows 8.

SweetLabs cofounder Chester Ng showed me the Zynga games on an Acer machine, and the arcade looks appealing. Users can install games in one click and start playing games immediately; the interface is pretty and everything loads very quickly. Most of all, the games have the feel of an authentic tablet experience.

Ng says the smart arcade features real time notification badges and banners, and starts tailoring games to what the user likes and doesn?t like.

?Pokki is trying to make the PC better for users, developers to help them get distribution, and now device manufacturers,? Ng tells me. ?There are plenty of problems to solve for all three of those constituencies.?

For developers like Zynga, Pokki offers a new distribution as they try to get their games into users hands beyond Facebook and mobile devices; these games are promoted right out of the box when a user opens a new PC. And it works for Pokki and equipment manufacturers, as Ng says it is a revenue opportunity for them to drive users to these games.

Pokki enables manufacturers like Acer to offer the newest, hottest games in real time, rather than preloading machines with a game months before the point of sale.

Ng said working with Acer is ?a big win for us,? and that it?s ?good validation that we?re not only making the pc better but also helping them on the business side.?

Malaysia declares emergency as Indonesia smoke pollution thickens

KUALA LUMPUR (Reuters) - Malaysia declared a state of emergency in two parts of the southern state of Johor on Sunday, as smoke from land-clearing fires in Indonesia pushed air pollution above the level considered hazardous.

The illegal burning of forests and other land on Indonesia's Sumatra island, to the west of peninsular Malaysia and Singapore, to clear space for palm oil plantations is a chronic problem during the June-September dry season.

The "haze" caused by fires in Riau province on Sumatra has also shrouded neighboring Singapore but air quality in the city state improved over the weekend after reaching hazardous levels.

"Prime Minister Najib Razak has agreed to declare emergency status in Muar and Ledang with immediate effect," Malaysian Natural Resources and Environment Minister G. Palanivel said in a Facebook post.

Palanivel said the air pollution index in the two districts had exceeded 750. A reading above 300 indicates that air pollution is hazardous.

Domestic media quoted the minister as saying cloud seeding would be carried out in the affected areas.

All 211 schools in the area are to be closed until further notice, residents have been advised to stay indoors and face masks have been distributed, Khaled Nordin, chief minister of the state, said, also via posts on Facebook.

Schools have been ordered shut in the neighboring state of Malacca, where pollution has also reached hazardous levels. Schools were also ordered to close in one district in Pahang state.

All domestic airports managed by Malaysia Airports Holdings Berhad (MAHB) are operating as usual despite the haze, the national news agency quoted the airport operator as saying.

The current visibility level of 1 km was still safe, but runways would have to close if visibility fell under 300 m, it quoted Malaysian airports official Azmi Murad as saying.

Indonesian officials have deflected blame by suggesting companies based in Malaysia and Singapore may be partly responsible. Malaysia-listed Sime Darby and Singapore's Wilmar Group both deny the charge.

EU finance ministers fail to agree on bank rules

LUXEMBOURG (AP) ? European Union finance ministers negotiating almost around the clock broke up unsuccessful talks on Saturday on how to downsize or close banks without letting taxpayers foot the bill and faced a danger that the divisive issue could undermine trust in Europe's ability to stabilize its financial system.

Irish Finance Minister Michael Noonan said that the negotiations he chairs would need another "full meeting" next Wednesday to bridge fundamental differences between the 27 member nations and warned "there is no guarantee it will reach conclusion."

Despite 19 hours of intricate negotiations, several ministers hinted at the prolonged impasse between the members of the 17-nation eurozone and the EU's ten other members like Britain that are not part of the currency union.

"It is principally an issue of the non-euro and the euro" nations, Noonan said.

French Finance Minister Pierre Moscovici voiced confidence that ministers will be able to broker a deal at the emergency meeting which comes only hours ahead of a summit of EU leaders to assess the brittle financial state of the union.

"I have no doubt we will reach a deal," Moscovici said.

An agreement on the rules would have been an important step to stabilizing Europe's financial system and establish a so-called banking union, which aims to give the supervision and rescue of banks to European institutions rather than leaving weaker member states to fend for themselves. It is a key part of the EU plans to restore financial and economic stability to the region.

The ministers at their meeting in Luxembourg sought to decide on new rules determining the order in which investors and creditors would have to pay for bank restructurings. A key stumbling block was who to hit hardest: Should losses be limited to banks' shareholders and creditors, or should small companies and ordinary savers holding uninsured deposits worth more than 100,000 euros ($132,000) also be included?

The most controversial issue, however, proved to be how much leeway member states should be granted in making decisions on winding down banks. Some countries like Britain don't want to be bound by rigid European rules. Other nations warned that too much flexibility would create new imbalances between the bloc's weaker and stronger economies and destroy the project of establishing a single set of rules that creates certainty for investors and restores trust in the financial system.

Moscovici said "90 percent of the work" was done, although France and others were still pushing for greater flexibility.

The ministers had vowed to resolve the issue by the end of June, thus the agreement on Wednesday's emergency meeting. Noonan however said the issue could spill beyond that, when Lithuania will take over the chairmanship of EU meetings.

Once the ministers finalize the legislation, they will then start negotiating the legislation with the European Parliament.

In addition to how much capital a bank must hold, the new European rules would also establish a minimum level of funds ? be it capital, bonds, or deposits ? that banks must have on their books to ensure that there's always enough privately held assets on which losses can be forced, thus shielding taxpayers from the burden of propping up the bank.

Following the 2008-2009 financial crisis, countries like Ireland, Britain and Germany each had to pump dozens of billions of fresh capital into ailing banks to avoid the financial system from collapsing.

Europe has already had to deal with problems involved in restructuring banks this year. Cyprus had to seek a rescue loan after it could no longer shoulder the cost of bailing out its banks.

An initial agreement with the island's European creditors and the International Monetary Fund sparked market fears since it exposed small savers with deposits under the 100,000 euro guarantee to losses.

The deal was rapidly overhauled, but holders of large deposits in some banks were forced to take harsh losses.

In the U.S., the Federal Deposit Insurance Corp.'s rules specify that deposits larger than $250,000 might have to take losses in case of bank failures, but Europe still lacks a common rule.

The new rules being discussed Friday will also foresee the establishment of national bank restructuring funds, which will eventually be merged into a European resolution authority, one of three planned parts of Europe's banking union.

Another part will be centralized oversight of big banks anchored at the European Central Bank due to be operational next year. But the discussion on the third section, a jointly guaranteed deposit insurance, is only in its early stages.

"The banking union is built brick by brick," Moscovici said Friday.

At their meeting, the finance ministers also rubber-stamped a seven-year extension of maturities on the bailout loans for Portugal and Ireland, granting the countries more financial leeway.

On Thursday, the finance ministers of the 17 EU countries that use the euro agreed on broad guidelines on how to use the bloc's permanent bailout fund to inject fresh capital into ailing banks as a means of last resort to keep banks from failing.

Enabling the 500 billion euro ($670 billion) rescue fund to shore up struggling banks directly is another long-promised building brick of the banking union.

(AP) ? A system-wide computer problem forced Southwest Airlines to ground its entire fleet of airplanes preparing for late-night departures, and cancellations were expected even after service slowly resumed early Saturday using a backup system, a company spokesman said.

Brad Hawkins told The Associated Press an estimated 250 flights were grounded at least temporarily due to the glitch, which impaired the airline's ability to do such things as conduct check-in, print boarding passes and monitor the weight of the aircraft.

Some flights were on the taxiway and diverted back to the terminal after the problem was detected around 11 p.m. ET Friday, he said. Flights already in the air were unaffected.

Hawkins said service resumed around 2 a.m. ET Saturday after officials began using a different system.

"Backup systems are in place not the main system, so it's slower," he said. "But we are able to start launching these flights."

He said at least some cancellations were expected because the airline doesn't do redeye flights and was near "the end of our operational day."

The late hour of the disruption meant the computer problem affected far more flights on the West Coast, but Hawkins said at least a few on the East Coast were grounded as well. Southwest, based in Dallas, conducts, on average, 3,400 flights a day.

A spokesman for Los Angeles International Airport said of about 25 inbound and outbound flights remaining Friday, only five departing flights were experiencing delays, of 30 to 80 minutes. At LA/Ontario International Airport (ONT), a total of three flights ? all departures ? were affected.

Four Southwest flights were temporarily held in Seattle, said Christina Faine, a Seattle-Tacoma International Airport spokeswoman.

One flight to Oakland, Calif., had been due to leave at 9:20 p.m. and departed before 11 p.m. Faine said late Friday night that an airport duty manager, Anthony Barnes, told her the others were expected to depart shortly.

Steve Johnson, a spokesman for Portland, Ore., International Airport, said he was not aware of any planes held up there.

On home turf show, Airbus steals Boeing's thunder

LE BOURGET, France (AP) ? Airbus beat its rival Boeing in the orders stakes at the Paris Air Show but both aircraft manufacturers were encouraged that their lucrative wide-body planes were finding new customers ? a sign that the global economy could be on the mend.

The show at Le Bourget, which alternates with the Farnborough Air Show on an annual basis, is a platform for the sales race between the world's two major aircraft makers, whose customers are increasingly interested in the new-generation, energy-efficient planes to offset huge increases in the price of jet fuel.

Though Europe's Airbus said Thursday it notched up about $70 billion in sales at the show, it acknowledged a problem with its large backlog, which will delay the delivery of jets. Boeing Co., which last week signaled it expected a lower tally than Airbus, announced orders worth $66 billion ? last year Boeing won the annual orders battle at Farnborough, south of London.

Each company tends to inflate its sales figures ? a common tactic is to notch up previously announced purchases that have been firmed up at the show. Customers routinely negotiate enormous discounts ? in at least one widely-publicized case, a markdown of more than 50 percent ? although the details are usually secret.

Though both companies announced a series of orders for their single-aisle jets, most interest at this year's event was centered on sales of their bigger planes. Longer-haul aircraft cost a lot more and have been in less demand over the past few years amid a tough global economic environment. However, along with a recovering global economy, there are signs that airliners are renewing their fleets.

"We continually keep underestimating how quickly Middle East, Far East and indeed emerging markets are growing. We're seeing a greater intensification of the mature nations realizing they have to invest in the future. That's why the A350 is doing well. That's why the 787 is doing well," said Howard Wheeldon, an aerospace analyst.

The number of passenger planes in the air is expected to double in the next 15 to 20 years, according to industry figures, including those released last week from Boeing. That's going to raise the pressure on airports already at capacity.

"You're not going to get into Charles de Gaulle 15 years from now with double the number of flights. We have to have larger aircraft," said John Leahy, Airbus' chief operating officer. "We'll be selling quite a few of them."

Airbus' new A350, which flew last week for the first time, notched 69 new orders ? including upgrades from earlier orders for older jets ? and the company also secured sales for 20 of its biggest aircraft, the A380.

A big order came from United Airlines, which agreed Thursday to buy 10 A350-1000s, the largest version of the jet which typically seats 350 passengers. The carrier also converted a previous order for 25 A350-900s into -1000s. The new orders are worth $3.3 billion at list prices.

The plane is still undergoing testing, but is expected to enter into service in the first half of next year. Because it has not yet logged enough flight hours, the jet is not on display at this week's show. But the company said it will do a fly-by on Friday over Le Bourget air field.

Chicago-based Boeing, meanwhile, officially launched its new 787-10 wide-body and claimed 102 orders for the aircraft, which like the A350 will make extensive use of lightweight carbon fiber. The company also scooped up orders for six of its 777s, another wide-body twin-engine.

"Time-slots are going to become more valuable," said Gerard Feldzer, an analyst who once led the Bourget Air and Space Museum. That will make the big planes more profitable, he added.

On the short-haul front, Airbus announced orders for 80 of its single-aisle jets Thursday. Leasing company Hong Kong Aviation Capital agreed to buy 40 of Airbus' new generation A320 jets and 20 new generation A321s.

Meanwhile, U.S.-based budget carrier Spirit Airlines scooped up 20 of the current generation A321s and also converted 10 previous orders for A320s into A321s. Spirit's planes will all be fitted with add-ons to the wings that will increase fuel efficiency and feature a single-class layout to let the airline squeeze in more passengers.

The fuel efficiency of Airbus' smaller jets is one of their major selling points. At list prices, the Hong Kong Aviation Capital deal is worth $6.3 billion, while Spirit's new orders are worth $2.1 billion.

Both the A320 and the A321 are on the smaller side; each seats about 180 passengers, though the A321 can be configured for up to 220.

Smaller jet purchases have made up the bulk of the orders this year as airlines renew their fleets or expand into new markets, notably in Asia and Latin America.

"The global economy is definitely looking a lot better than it was a year ago," said Wheeldon, the analyst. "Clearly Airbus is leaps and bounds in front of Boeing in terms of numbers. We need two very healthy aircraft manufacturers in the Western world. ... We need them to be competitive against each other. We need that competition to drive technology."

'Voice' finalist disagrees with coach's pick

In their final performance show on Monday night, ?Voice? finalists Danielle Bradbery, Michelle Chamuel and The Swon Brothers were asked to reprise songs that their coaches chose as their ?defining moment.?

After their repeat renditions, the artists told TODAY.com if they agreed with their coaches? picks, and why those tracks might have stood out from all the others they?ve performed over the six weeks of live playoffs.

For teenage fan favorite Danielle, Blake Shelton chose the Pam Tillis song ?Maybe It Was Memphis,? and Danielle told us that ?I agreed with that choice. That was the song that went up so high on iTunes and everyone was freaking out. It's such a fun performance. It's so powerful. I love that song.?

She added that she?s been watching her earlier ?Voice? clips to see how far she?s come since starting on the show. ?I've watched my blind auditions, knockouts, battles, those performances that were way at the beginning,? she said.

?Just the clips that they showed for the (top 16 group) performance -- it's crazy how much my voice has changed and how little I look!? Danielle laughed, before clarifying, ?I'm still little, but I feel like I grew up a lot.?

Michelle wasn?t sure why Usher picked Taylor Swift?s ?I Knew You Were Trouble,? which she first performed only two weeks ago.

?I don't fully know,? she admitted. ?I think it's just the energy. That was one of the most unique (performances) for me as far as what I'm able to show. This is like the top of the energy chain for me. It's like, 'Let's turn it back up to 11.??

She revealed that it wasn?t the track she would?ve chosen as her defining moment. ?I very much respect his choice,? she continued, ?(but) I would've picked ?Grenade,? because that was the first time I felt all these interlocking pieces come together -- the score, the movement. That is what I would've picked.?

Of the decision to go back to their version of Loggins and Messina?s ?Danny?s Song,? Colton Swon of The Swon Brothers told TODAY, ?That was all (Blake). That?s our coach and we have faith in him.?

?Danny?s Song? wasn?t the first time America really took notice of the Swon Brothers. They also received universal praise when they performed George Jones? ?Who?s Gonna Fill Their Shoes? in tribute to the late country legend. So why didn?t they go with Jones instead?

?I think ('Danny?s Song') reached a bigger audience than the George Jones song did,? Zach Swon explained. ?It went No. 2 overall on the iTunes charts. I think it connected with a lot more people that may not be hardcore country fans. If you'd told me that was going to be our defining moment at the beginning of the season, I'd have called you crazy. But looking back on it now and all the feedback we got, I really think it bumped us up there.?

One of these three acts is about to have another defining moment. ?The Voice? will name its season four winner with a special two-hour Tuesday show beginning at 9 PM on NBC.

Protein essential for normal heart function identified

June 17, 2013 ? A study by researchers at Skaggs School of Pharmacy and Pharmaceutical Sciences and the Department of Pharmacology at the University of California, San Diego, shows that a protein called MCL-1, which promotes cell survival, is essential for normal heart function.

Their study, published in the June 15 online issue of the journal Genes & Development, found that deletion of the gene encoding MCL-1 in adult mouse hearts led to rapid heart failure within two weeks, and death within a month.

MCL-1 (myeloid cell leukemia-1) is an anti-apoptotic protein, meaning that it prevents or delays the death of a cell. It is also a member of the BCL-2 family of proteins that regulate mitochondria -- the cell's power producers -- and cell death. Aberrant expression of anti-apoptotic BCL-2 family members is one of the defining features of cancer cells, and is strongly associated with resistance to current therapies. Thus, these proteins are currently major targets in the development of new therapies for patients with cancer.

But, while MCL-1 is up regulated in a number of human cancers, contributing to the overgrowth of cancer cells, it is found at high levels in normal heart tissue. Additionally, the researchers found that autophagy -- a process which deals with mitochondrial maintenance and is normally induced by myocardial stress -- was impaired in mice with MCL-1 deficient hearts.

In summary, the study demonstrated that the loss of MCL-1 led to rapid dysfunction of mitochondria, impaired autophagy and heart failure, even in the absence of cardiac stress.

"Cardiac injury, such as a heart attack, causes levels of MCL-1 to drop in the heart, and this process may increase cardiac cell death," said ?sa B. Gustafsson, PhD, an associate professor at UCSD Skaggs School of Pharmacy and Pharmaceutical Sciences. "Therefore, preserving normal levels of this protein in cardiac tissue could reduce damage after a heart attack and prevent progression to heart failure."

By compromising both autophagy and mitochondrial function, MCL-1 inhibitors are likely to affect the cells' energy supply. "Our findings raise concerns about the potential cardiac toxicity of drugs that block MCL-1 -- drugs that have entered clinical trials because they increase cancer cell death," said the study's first author, Robert L. Thomas.

Galaxy acquire midfielder Mastroeni from Rapids

CARSON, Calif. (AP) - The Los Angeles Galaxy have acquired six-time MLS All-Star midfielder Pablo Mastroeni in a trade with the Colorado Rapids.

Under the deal announced Monday, the Galaxy also received the rights to midfielder Baggio Husidic. The Rapids received an international roster slot through the end of the 2014 MLS season and Los Angeles' second-round pick in the 2014 MLS SuperDraft.

Mastroeni will be eligible to make his Galaxy debut when Los Angeles hosts the Portland Timbers on Wednesday night.

The 36-year-old Mastroeni has spent the last 12-plus seasons with the Rapids, leading the club to its first MLS Cup championship in 2010.

A former member of the U.S. men's team, Mastroeni played in the 2002 and 2006 World Cup under current Galaxy coach Bruce Arena.

Obama taps finance director as envoy to Denmark

WASHINGTON (AP) ? Handing plum European posts to key campaign boosters, President Barack Obama on Friday nominated his former finance director to be U.S. ambassador to Denmark and tapped two major fundraisers for postings in Spain and Germany.

Fundraising guru Rufus Gifford, who raised upward of $700 million as the head of Obama's 2012 finance operation and held a similar job previously at the Democratic National Committee, is Obama's pick for Denmark, the White House said. Obama selected James Costos, a vice president of HBO, as his envoy to Spain.

If confirmed by the Senate, Gifford and Costos would bolster the limited ranks of ambassadors in U.S. history who have been openly gay. When Obama took office in 2009, only two previous ambassadors had been openly gay. Friday's nominations mark the fifth and sixth, gay rights groups said.

The Gay & Lesbian Victory Fund, which works to elect openly gay candidates, hailed the nominations and said they would clear the way for more gay Americans to achieve prominent positions in public service.

Obama also said he had selected John Emerson, an investment manager and former White House official, as his envoy to Germany. Both Costos and Emerson were listed in public disclosures as bundlers, meaning they raised more than $500,000 from others to benefit Obama's campaign. Federal Election Commission records show Costos donated more than $67,000 to efforts to re-elect Obama in 2012.

Ken Hackett, who stepped down last year as the president and CEO of Catholic Relief Services, is Obama's pick to be the U.S. emissary to the Vatican. Hackett worked extensively in Africa coordinating Catholic charitable programs on the continent.

Patricia Marie Haslach, a veteran diplomat who has worked in Iraq, Pakistan and Laos, is Obama's pick to be ambassador to Ethiopia. He nominated Liliana Ayalde, another State Department official with experience throughout Latin and Central America, for the top post in Brazil. Both Haslach and Ayalde have held the rank of ambassador before.
